Marisa Tomei played the role of May, Peter Parker’s aunt, since Spider-Man’s introduction to the MCU in 2016’s Captain America: Civil War. Since then, she’s reprised the role for multiple Spider-Man movies until her death in Spider-Man: No Way Home.
There is no official confirmation for whether Marisa Tomei will be reprising her role for the upcoming Spider-Man: Brand New Day film, whether in a small or big capacity. So far, we don’t know too much about the movie aside from the one trailer that’s been dropped. This does not mean we can discount her appearance in the upcoming film due to the following reasons.

Spider-Man: Brand New Day will be Peter Parker’s darkest arc yet, following the death of his beloved Aunt May at the hands of Green Goblin. Marisa Tomei has not appeared in the trailer for the film, but a graveyard scene indicates that Peter Parker continues to show his love.
The gravestone indicated that she died at the age of 60, and she still could appear in flashback sequences never seen before in Peter Parker’s life. We’ve seen Peter Parker document a lot of his life on his phone, like in Spider-Man: Homecoming, opening up the chance that we “May” see multiple footage of her on Peter’s phone.
The webcrawler is going through a metamorphosis as its genes mutate, opening up the possibilities for hallucinations as well. The story will revolve around this evolution, along with Peter’s isolation following No Way Home. With nobody remembering him and his Aunt gone, this gives a lot of time for the character to introspect or be stuck in the past: something we will get to see on the big screen.

The events of Far From Home dealt heavily with Peter’s grief and how he will continue Stark’s legacy, and he wishes to be a superhero looking out for the little guy.

The only family he had left is gone, giving him no support system for when things go really wrong. The Sam Raimi films showed just how important Aunt May was, with Peter regularly visiting her when things were at an all-time low. He could always rely on her to dispense life-changing wisdom with just simple words.
The fear of losing another loved one will drive Spider-Man/Peter Parker into further isolation as he moves into a new, frightening chapter where he isn’t in control of his body.
